Product Specific Information

The monoclonal antibody TWAJ recognizes mouse and human Gata-3, a T cell-specific transcription factor crucial for thymic development and Th2 differentiation.
Gata-3 expression is found in the CNS, skin, mammary glands, and kidney during embryonic development. Homozygous knockout of Gata-3 is embryonic lethal, highlighting its essential role.

In the thymus, Gata-3 expression is mainly on CD4 single-positive cells. During Th2 differentiation, Gata-3 binds to the IL-4 promoter and represses T-bet expression, inhibiting Th1 differentiation.
Alternative splice variants (50 and 45 kDa) are recognized by this antibody, including those found in the MCF7 cell line.

Staining Requirements: Use with Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
Crossreactivity: Published in rhesus monkey.

• Intracellular staining followed by flow cytometric analysis.

Applications Tested

• Pre-diluted and tested by intracellular staining of mouse thymocytes using Foxp3/Transcription Factor Staining Buffer Set and protocol.
• Recommended use: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information

Brilliant Violet™ 711 (BV711) emits at 713 nm and is suitable for cytometers with a violet (405 nm) laser.
Use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions when using multiple polymer dyes.

• Light Sensitivity: Protect from light to prevent photo-induced oxidation.
• Fixation: Samples can be stored in IC Fixation Buffer or 1-step Fix/Lyse Solution up to 3 days at 4°C in the dark.
• Compatibility: BV711 is not compatible with methanol-based fixation.
• Excitation: 407 nm; Emission: 713 nm; Laser: Violet laser.

Target Information

GATA3 regulates T-cell antigen receptor (TCR) subunit genes (alpha, beta, gamma, delta) through enhancer-binding activity.
It contains zinc finger domains and is expressed in T cells but not B cells or macrophages.
The human GATA3 gene spans 17 kb with six exons; its zinc finger regions are highly conserved with GATA1.
