Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45, also known as Leukocyte Common Antigen (LCA). CD45 is expressed by all hematopoietic cells except circulating erythrocytes and platelets, and plays a critical role in lymphocyte activation through its tyrosine phosphatase activity.

• Applications Tested: Pre-diluted and validated for flow cytometry of normal human peripheral blood cells
• Recommended Use: 5 µL (0.5 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test)

Brilliant Violet™ 711 (BV711) emits at 713 nm and is optimized for cytometers equipped with a 405 nm violet laser. Ensure compatibility before use.

Usage Notes

• When using multiple polymer dye-conjugated antibodies (e.g., Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to reduce non-specific interactions.
• Light sensitivity: Protect from light to prevent photo-oxidation.
• Fixation: Samples can be stored in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) for up to 3 days at 4°C in the dark.
  Note: BV711 is not compatible with methanol-based fixation.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It is absent from non-hematopoietic cells and tissues. CD45 is essential for lymphocyte development and antigen receptor signaling, regulating Src-family kinases and JAK kinase activity. Isoforms of CD45 arise from alternative splicing and differ in extracellular domains, influencing cell-type-specific expression and function.
