Product Specific Information

The monoclonal antibody OX104 recognizes human CD200 (OX2), a member of the Ig superfamily with two Ig domains, a transmembrane and cytoplasmic domain. CD200 is expressed on resting and activated B and T cells, keratinocytes, peripheral and central nerve cells, follicular dendritic cells, and ovarian cells. Interaction with CD200R results in macrophage activation (IL-6 production), inhibition of mast cell degranulation, reduced TNF-α and IL-13 secretion, and overall attenuation of lymphocyte activation. Expression on trophoblasts suggests a role in maternal tolerance.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested with normal human peripheral blood cells at 5 µL (0.5 µg) per test. A test corresponds to staining a 100 µL cell sample (10⁵–10⁸ cells/test).

Fixation:
Samples may be stored in IC Fixation Buffer or 1-step Fix/Lyse Solution for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer or Brilliant Stain Buffer to minimize non-specific interactions.

Target Information

CD200 is a type-1 membrane glycoprotein containing two immunoglobulin domains, belonging to the immunoglobulin superfamily. It interacts with CD200R on myeloid cells to prevent macrophage activation. Homologues exist in herpesviruses and poxviruses. CD200 may regulate myeloid activity and inhibit macrophage lineage activation. Multiple alternatively spliced transcript variants exist.
Diseases associated with CD200 dysfunction include Mantle Cell Lymphoma and Primary Hypertrophic Osteoarthropathy.
