Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is essential for assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells. Crosslinking of TCR via immobilized UCHT1 induces intracellular activation and proliferation.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of normal human peripheral blood cells.
Use ≤ 5 µL (0.4 µg)/test in 100 µL final volume containing 10^5–10^8 cells. Optimize antibody titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) enhances staining performance and should be used with all NovaFluor conjugates.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10^3–10^8 cells).

NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures designed to minimize spillover and spread.

Target Information

The CD3 subunit complex transduces antigen-recognition signals in T cells and regulates TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are structurally related members of the immunoglobulin superfamily encoded on chromosome 11. They associate with cytoplasmic signal transduction molecules via double tyrosine-based motifs.

CD3 antigen is found on:

• 68–82% of normal peripheral blood lymphocytes
• 65–85% of thymocytes
• Purkinje cells in the cerebellum

It is not expressed on B or NK cells. Decreased T lymphocyte percentages may occur in autoimmune diseases. Defects in the CD3 gene are linked to CD3 immunodeficiency.
