Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta subunits, is required for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Crosslinking via immobilized UCHT1 induces cellular activation and proliferation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells (5 µL/test). A test corresponds to staining a cell sample in 100 µL volume with 10⁵–10⁸ cells/test.

NovaFluor dye compatibility:
Not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

CellBlox Plus Blocking Buffer:
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample.

NovaFluor Technology:
Based on Phiton technology utilizing nucleic acid dye structures for engineered fluorescent signatures with minimal spillover.
Excitation: 509 nm; Emission: 665 nm; Laser: 488 nm (Blue).

Target Information

The CD3 complex transduces antigen-recognition signals in T cells and regulates TCR surface expression. It comprises CD3 gamma, delta, epsilon, and zeta subunits, encoded on chromosome 11. These subunits contain cytoplasmic tails with tyrosine-based motifs mediating signal transduction. CD3 is expressed on most T lymphocytes and thymocytes but not on B or NK cells. Defects in CD3 genes are associated with immunodeficiency.
