Description:
The UCHT1 monoclonal antibody reacts with human CD3ε, a 20 kDa subunit of the TCR complex. Together with CD3γ and CD3δ, the ε chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells. Crosslinking via UCHT1 induces intracellular signaling leading to activation and proliferation.
Applications Reported:
Flow cytometric analysis.
Applications Tested:
Pre-titrated and tested for flow cytometric analysis of normal human peripheral blood cells. Use 5 µL (0.8 µg) per test in 100 µL final volume (10⁵–10⁸ cells/test).
Kit Contents:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Usage Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability detection.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample to antibody cocktails before combining with cells.
- For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates utilize Phiton™ technology, employing novel nucleic acid dye structures for engineered fluorescence with minimal spillover and spread.

Target Information
The CD3 subunit complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It consists of CD3γ, CD3δ, CD3ε, and CD3ζ subunits, encoded on chromosome 11. These subunits have cytoplasmic tails that associate with signal transduction molecules via tyrosine-based motifs.
CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes but not on B or NK cells. Defects in CD3 genes can lead to immunodeficiency.
