Product Specific Information

Description:
The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed on thymocytes and subsets of mature T and NK cells. CD8 binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T-cell development and activation.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested for flow cytometry on normal human peripheral blood cells. Recommended usage is 5 µL (0.8 µg) per test in a final volume of 100 µL.

Compatibility Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for optimal results.

Blocking Buffer Recommendation:
This conjugate ships with CellBlox Plus Blocking Buffer (Product # C001T06F01), which should be used for all experiments involving NovaFluor conjugates.

  • Add 5 µL per sample (regardless of number of NovaFluor dyes).
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures that provide engineered fluorescent signatures designed to minimize spillover and spread.

492 nm / 766 nm / 488 nm (Blue laser)

Target Information

The CD3 subunit complex transduces antigen-recognition signals into T cells and regulates TCR complex expression. CD3 subunits (gamma, delta, epsilon, zeta) are structurally related immunoglobulin superfamily members encoded on chromosome 11. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes and thymocytes but not on B or NK cells. Defects in CD3 genes are associated with CD3 immunodeficiency.
