Crk-I와 Crk-II 형태는 생물학적 활성에서 차이를 보인다. Crk-II는 Crk-I보다 낮은 변형 활성을 가지며, Rac 의존적 MAPK8 활성화, 막 주름 형성, 세포 이동에 관여한다. 또한 DOCK1 및 DOCK4와의 상호작용을 통해 세포사멸세포의 식세포작용 및 세포 이동을 조절할 수 있으며, EFNA5-EPHA3 신호 조절에도 관여할 수 있다.
