Target Information
ERN1 (serine/threonine kinase), also known as endoplasmic reticulum to nucleus signaling 1 (IRE1), is a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities, playing a key role in ER stress signaling. ERN1 regulates IRE1 proteolysis in mammalian cells and is essential for fetal viability in extraembryonic cells. IRE1 senses unfolded proteins in the ER and initiates the unfolded protein response (UPR) by splicing XBP1 mRNA, converting it into a potent transcriptional activator. Although null mutants are viable, IRE1 is essential under ER stress conditions. Depletion of IRE1p has no effect under normal conditions but induces apoptosis under stress.
