Product Specific Information
The OKT9 monoclonal antibody reacts with human CD71, a 170–180 kDa type II transmembrane protein known as the transferrin receptor. CD71 exists as a homodimer and is essential for cellular growth. It is expressed by immature proliferating cells and at low levels on resting mature lymphocytes. Upon antigen or mitogen stimulation, T and B cells upregulate CD71 expression.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated for flow cytometry using PHA-stimulated human PBMCs. Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.
Excitation: 488 nm; Emission: 520 nm; Laser: Blue.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Cellular iron uptake occurs via receptor-mediated endocytosis of transferrin-bound CD71 into endosomes, where acidification releases iron. The apotransferrin-receptor complex recycles to the cell surface, dissociating at neutral pH. Transferrin receptor is crucial for erythrocyte and nervous system development. Serum transferrin receptor (sTfR) is used to detect EPO misuse and diagnose anemia associated with conditions such as rheumatoid arthritis, pregnancy, irritable bowel syndrome, and HIV.
