Target Information

Activator of S phase kinase (ASK, DBF4) is a regulatory subunit of the human Cdc7 kinase complex that plays a pivotal role in the G1/S cell cycle transition. Elevated ASK expression in late G1 contributes to increased Cdc7 kinase activity at the G1/S boundary. ASK interacts with replication origins, suggesting that Cdc7 may trigger S phase by activating replication initiation complexes. ASK and Cdc7 are involved in regulating DNA replication initiation by targeting MCM2 protein. The nuclear localization and chromatin binding of ASK increase at late G1 phase, potentially requiring additional signals. ASK may also functionally interact with menin, a tumor suppressor mutated in MEN1, in cell proliferation regulation.
