Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 upregulates genes like p21waf1 involved in G1 or G2/M arrest. Structurally, it includes an N-terminal transactivation domain, central DNA-binding domain, oligomerisation domain, and C-terminal regulatory domain.

Phosphorylation at Ser15 is critical for activation and stabilization, while Ser392 phosphorylation is associated with tumor formation. p53 plays key roles in preventing proliferation of damaged cells and acts as an anticancer factor. It is also linked to aging, oxidative stress, and cognitive processes such as long-term potentiation (LTP). Normally expressed at low levels, p53 is overexpressed in transformed cell lines, contributing to malignancy. Mutations in TP53 gene cause loss of tumor suppressor function and are observed in many cancers, including hereditary cases such as Li-Fraumeni syndrome.
