Target Information
Furin is a membrane-associated, calcium-dependent serine protease belonging to the subtilisin-like prohormone convertase (PC) family. Members of this family cleave most prohormones and neuropeptide precursors, as well as various viral and bacterial proteins.
Furin and related PCs share structural features including:
- ~10 kDa amino-terminal proregion
- ~55 kDa conserved catalytic domain
- Variable-length carboxyl-terminal domain
Furin becomes active after proregion cleavage and is unique among PCs for having a transmembrane domain. It cleaves substrates at the carboxyl-terminus of the consensus sequence RX(K/R)R.
The peptide sequence C771PSDSEEDEG780 localizes furin to the trans-Golgi network, and phosphorylation within this region regulates its intracellular routing. The Y759KGL762 sequence mediates internalization from the cell surface.
