Product Specific Information
This AGO2 11A9 monoclonal antibody recognizes human Argonaut 2 (Ago2), a transcriptional regulator involved in RNA interference and endonucleolytic cleavage.
- Detects reduced Ago2 by Western Blot.
- Stains Ago2 in cytoplasm and nucleus in Immunocytochemistry.
- Validated for Western Blot and Immunocytochemistry/Immunofluorescence.
- Does not cross-react with murine Ago2.
- Tested on human cancer cell lysates and fixed human U-2 OS cells.
- Recommended concentrations: ≤4 µg/mL for WB and ≤1.25 µg/mL for ICC/IF.
Target Information
Ago proteins are broadly expressed in somatic cells and associate with miRNAs, playing key roles in RNA silencing pathways.
Only Ago2 exhibits endonucleolytic “Slicer” activity, enabling miRNA-directed mRNA cleavage when base-pairing is perfect.
In cases of partial complementarity, Ago2 represses translation instead of cleavage.
Ago2 is essential for:
- RNA interference and translational repression
- Heterochromatinization and small RNA-guided gene silencing
- mRNA degradation via RISC complex
- miRNA maturation regulation
Overexpression of Ago2 correlates with tumor growth and survival in cancer patients.
Mouse gene disruption studies show Ago2 is crucial for embryonic, B-lymphoid, and erythroid development.
