This EP10 monoclonal antibody reacts with human Synaptophysin (Major Synaptic Vesicle Protein p38), a 38-kDa integral membrane protein expressed on neuronal presynaptic vesicles in the brain, spinal cord, retina, neuromuscular junctions, and adrenal medulla. It is also expressed in neuroendocrine tumors and neuroblastomas and plays a role in synaptic vesicle trafficking.

The EP10 antibody recognizes human and hamster synaptophysin but not mouse or rat synaptophysin.

Applications Reported: Immunoprecipitation, Western blotting (WB), immunohistochemical staining (formalin-fixed paraffin embedded tissue), and immunocytochemistry (ICC).

Applications Tested:

• IHC on formalin-fixed paraffin-embedded human cerebellum using citrate buffer antigen retrieval (≤5 µg/mL).
• WB on human brain lysate (≤5 µg/mL).
  It is recommended to titrate the antibody for optimal assay performance.

Target Information

Synaptophysin is a calcium-binding integral membrane glycoprotein present in presynaptic vesicles of nearly all neurons. It has four transmembrane domains and forms a complex with dynamin at high calcium concentrations, suggesting a role in synaptic vesicle endocytosis. It is involved in short-term and long-term synaptic plasticity and is widely used as a marker for nerve terminals and neuroendocrine tumor differentiation. Mutations in this gene can lead to X-linked mental retardation.
