Product Specific Information
This AFP3 monoclonal antibody reacts with human alpha-fetoprotein (AFP), a 70-kDa secretory protein of the albumin gene family. AFP is synthesized by the yolk sac and fetal liver during embryogenesis, with high levels in fetal serum that decrease dramatically after birth. Elevated AFP levels in adults are associated with hepatocellular carcinoma, germ cell teratoblastoma, liver regeneration, viral hepatitis, and cirrhosis, making AFP detection a diagnostic marker for these conditions.
For western blotting or immunohistochemistry on paraffin sections, use monoclonal antibody 1E8 (Product # 14-9499).
Applications Reported: Immunoblotting (WB) and Immunocytochemistry (ICC).
Applications Tested:
- Western blot analysis of reduced HepG2 cell lysate (1–10 µg/mL).
- Non-reduced lysate yields a band at ~58 kDa.
- Immunocytochemistry of HepG2 cells (1–5 µg/mL).
- Antibody should be titrated for optimal performance.

Target Information
AFP (Alpha Fetoprotein) is a major plasma protein produced by the yolk sac, intestinal tract, and liver during fetal life. In adults, AFP expression is associated with hepatoma or teratoma, but hereditary persistence may occur without pathology. AFP is considered the fetal counterpart of serum albumin, encoded in tandem with the albumin gene on chromosome 4. It exists in monomeric, dimeric, and trimeric forms and can bind copper, nickel, fatty acids, and bilirubin. AFP levels in amniotic fluid are used to screen for spina bifida and anencephaly. Elevated serum AFP is observed in hepatocellular carcinoma, teratoblastoma, colorectal, pancreatic cancers, and germ cell neoplasms.
