Target Information
This gene encodes a member of the F-box protein family characterized by an approximately 40 amino acid motif, the F-box. F-box proteins are part of the SCF ubiquitin ligase complex (SKP1-cullin-F-box), functioning in phosphorylation-dependent ubiquitination.
F-box proteins are divided into three classes:
- Fbws with WD-40 domains
- Fbls with leucine-rich repeats
- Fbxs with other protein-protein interaction modules or no recognizable motifs
The encoded protein belongs to the Fbws class and contains multiple WD-40 repeats. It is homologous to Xenopus bTrCP1, yeast Met30, Neurospora Scon2, and Drosophila Slimb proteins. It interacts with HIV-1 Vpu, connecting CD4 to the proteolytic machinery, and associates with phosphorylated IkappaBalpha and beta-catenin destruction motifs.
This protein likely functions in multiple transcriptional programs by activating the NF-kappaB pathway and inhibiting the beta-catenin pathway.
