Target Information
Protein tyrosine phosphatases (PTPs) are enzymes that remove phosphate groups from phosphorylated tyrosine residues on proteins. Together with tyrosine kinases, PTPs regulate phosphorylation of key signaling molecules such as the MAP kinase family.
Dual specificity phosphatases (DSPs) are a subclass of PTPs that selectively dephosphorylate phosphothreonine and phosphotyrosine residues. The prototypical DSP is the VH1 gene in vaccinia virus expressed in late-stage infection.
VHR’s shallow active site allows hydrolysis of phosphorylated serine, threonine, or tyrosine residues, while deeper active sites in PTPs restrict substrate specificity to phosphotyrosine only.
