Product Specific Information

The SIDI8BEE monoclonal antibody reacts with human and rhesus macaque CD8 beta. The CD8b chain associates with the CD8a chain to form the CD8αβ heterodimer expressed on most thymocytes and peripheral cytotoxic αβ T cells. The CD8 receptor binds MHC class I and plays a key role in T cell development and activation.

SIDI8BEE detects CD8β in both CD8αβ heterodimers and CD8ββ homodimers. Cross-reactive with human and rhesus macaque CD8β; other primate species not tested.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry on human peripheral blood lymphocytes. Use 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

The CD8B antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes. It functions as a co-receptor with the T-cell receptor, recognizing antigens presented by MHC class I molecules. CD8 can form either αα homodimers or αβ heterodimers, both structurally similar to immunoglobulin light chains. The CD8B gene encodes the beta chain isoforms, with multiple alternatively spliced variants and a pseudogene located on chromosome 2.
