Product Specific Information

The SIDI8BEE monoclonal antibody reacts with human and rhesus macaque CD8 beta.
CD8β associates with CD8α to form the CD8αβ heterodimer expressed on thymocytes and peripheral cytotoxic T cells.
The CD8 receptor complex binds to MHC class I and plays a role in T cell development and activation.
SIDI8BEE detects CD8β in both CD8αβ heterodimers and CD8ββ homodimers.
Cross-reactive to human and rhesus macaque CD8β; other primate species not tested.

Each product includes:

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry of human peripheral blood lymphocytes (5 µL per test).
A test is defined as the antibody amount that stains cells in a 100 µL final volume (10⁵–10⁸ cells/test).

NovaFluor dye compatibility:
Not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample or per 100 µL of cell sample for single-color controls.

Technology:
NovaFluor conjugates are based on Phiton™ technology using engineered nucleic acid dye structures for optimized fluorescence performance (Excitation: 509 nm; Emission: 614 nm; Laser: 488 nm).

Target Information

The CD8B antigen is a cell surface glycoprotein found on cytotoxic T lymphocytes.
Acts as a coreceptor with the T-cell receptor recognizing antigens presented by MHC class I molecules.
Functional forms include:

• Homodimer (αα)
• Heterodimer (αβ)

Both chains share homology with immunoglobulin variable light chains.
CD8B gene encodes beta chain isoforms; multiple spliced variants and a pseudogene exist on chromosome 2.
