Product Specific Information

The SIDI8BEE monoclonal antibody reacts with human and rhesus macaque CD8 beta.
CD8b chain associates with CD8a to form the CD8ab heterodimer expressed on thymocytes and peripheral cytotoxic αβ T cells.
The CD8 receptor complex binds to MHC class I and plays a role in T cell development and activation.
SIDI8BEE detects CD8β in both CD8ab heterodimer and CD8bb homodimer and is cross-reactive to human and rhesus macaque CD8b.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested using human peripheral blood lymphocytes; use at 5 µL (0.8 µg) per test.
A test = amount of antibody staining a sample in 100 µL volume (10⁵–10⁸ cells/test).

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01)

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample before combining with cells
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells)

Target Information

CD8B antigen is a cell surface glycoprotein on cytotoxic T lymphocytes mediating cell-cell interactions.
Acts as a coreceptor with the T-cell receptor, recognizing antigens presented by class I MHC molecules.
Functional forms include homodimers (αα) or heterodimers (αβ).
Both chains share homology with immunoglobulin variable light chains.
This gene encodes CD8β isoforms; multiple alternatively spliced variants exist.
A pseudogene is also located on chromosome 2.
