The 56 kDa protein ACAT1 (acetyl-CoA acetyltransferase 1) is primarily involved in cellular cholesterol homeostasis. It belongs to the membrane-bound O-acyltransferase family and is present in various cell types. In mammals, ACAT1 exists in two isoenzyme forms: ACAT1 and ACAT2. It is composed of nine transmembrane domains (TMDs) with two active sites — His460 located within TMD7 and Asn421 located within the fourth large cytoplasmic loop.
