Product Specific Information

The monoclonal antibody TWAJ recognizes mouse and human Gata-3, a member of the Gata transcription factor family.
Gata-3 is a T cell-specific transcription factor crucial for thymic development and Th2 differentiation.
Expression during embryogenesis occurs in the central nervous system, skin, mammary glands, and kidney.
Homozygous knockout of Gata-3 is embryonic lethal, confirming its essential role.

In thymus, expression is mainly found in CD4 single positive cells.
During Th2 differentiation, Gata-3 binds to the IL-4 promoter and represses T-bet expression, thus inhibiting Th1 differentiation.

Alternative splice variants (50 and 45 kDa) have been reported, notably in MCF7 cells.
The TWAJ antibody recognizes both forms.

For staining, use the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
Cross-reactivity in rhesus monkeys has been published.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and tested on mouse thymocytes using the Foxp3/Transcription Factor Staining Buffer Set.
Recommended usage: 5 µL (0.5 µg) per test (100 µL final volume). Cell number may vary between 10⁵–10⁸ cells/test.

Brilliant Ultra Violet™ 395 (BUV395) emits at 395 nm and is optimized for UV (355 nm) laser-equipped cytometers.
Ensure instrument compatibility before use.

When using multiple polymer dye-conjugated antibodies (e.g., Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Target Information

GATA3 regulates transcription of T-cell receptor (TCR) subunits (alpha, beta, gamma, delta) by binding enhancer elements.
It contains zinc finger domains and is expressed in T cells but not B cells or macrophages.
Human GATA3 gene has 6 exons over 17 kb, with high conservation to GATA1 in zinc finger regions.
GATA3 plays a pivotal role in T-cell lineage commitment and enhancer-mediated activation.
