Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein belonging to the immunoglobulin receptor family. It plays a key role in signal transduction, T cell differentiation, and activation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer (CD8α and CD8β) and functions as a co-receptor with the T cell receptor (TCR) for MHC class I molecules. CD8α is essential for MHC-I binding. It can also be expressed as CD8α homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Interaction with MHC-I/peptide complexes triggers Lck kinase recruitment, leading to cytokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are crucial for pathogen and tumor cell clearance. IL-2, IL-12, and TGF-β1 strongly enhance the differentiation of naïve CD8+ T cells into CTLs.
