Product Specific Information

FACS procedure using PBMCs

  • Add 10 µL of MA1-12030 per 10⁶ PBMC in 100 µL PBS.
  • Mix gently and incubate for 15 minutes on ice.
  • Wash twice with PBS and analyze, or fix with 0.5% v/v paraformaldehyde in PBS before analysis.

FACS procedure using whole blood

  • Add 10 µL of MA1-12030 per 100 µL of whole blood.
  • Mix gently and incubate for 15 minutes at room temperature.
  • Lyse the whole blood, wash once with PBS, and analyze or fix with 0.5% v/v paraformaldehyde in PBS before analysis.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family that plays an essential role in signal transduction, T cell differentiation, and activation.

  • Expressed mainly on T cells as a disulfide-linked heterodimer of CD8α and CD8β.
  • Functions as a co-receptor with TCR for MHC class I molecules.
  • CD8α chain is critical for MHC-I binding.
  • Also found on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers.
  • Engagement of MHC-I/peptide complexes triggers Lck recruitment, leading to lymphokine production and CTL activation.
  • Activated CTLs are essential for pathogen and tumor cell clearance.
  • Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
