Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein belonging to the immunoglobulin family of receptors. It plays a critical role in signal transduction, T cell differentiation, and activation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with the T cell receptor (TCR) for MHC class I molecules. In contrast, CD4 acts as a co-receptor for MHC class II molecules.

CD8α is essential for MHC-I binding and is also expressed as a homodimer on subsets of T cells, NK cells, monocytes, and dendritic cells. Engagement of MHC-I/peptide complexes by antigen-presenting cells (APCs) recruits Lck kinase, leading to cytokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are crucial for pathogen and tumor cell clearance. Differentiation of naïve CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
