Target Information

Janus kinase 2 (JAK2) is a protein tyrosine kinase involved in cytokine receptor signaling pathways and is often mutated in polycythemia vera. It contains the catalytic domain (JH1) of JAK2, and its activation leads to phosphorylation of STAT proteins, which then dimerize and translocate to the nucleus to regulate transcription of interferon-stimulated genes.
JAK2 is essential for IFN gamma-receptor complex initiation, with JAK1 acting as an amplifier. Mutations in JAK2 are associated with diseases such as Budd-Chiari syndrome, Polycythemia vera, Thrombocythemia 3, myelofibrosis, and Acute myelogenous leukemia.
