This antibody detects endogenous levels of Nucleophosmin only when phosphorylated at Ser4.
Target Information
NPM1 (Nucleophosmin 1, also known as B23, nutramin, NO38) is a ubiquitously expressed phosphoprotein involved in multiple cellular processes, including:
- Ribosome assembly and transport
- Cytoplasmic/nuclear trafficking
- Regulation of DNA polymerase alpha activity
- Centrosome duplication
- p53 regulation
NPM1 continuously shuttles between the nucleus, cytoplasm, and nucleolus, acting as a chaperone for core histones. It binds nucleic acids, prevents protein aggregation, and facilitates protein renaturation after denaturation. NPM1 is associated with centrosome duplication and is a substrate of CDK2/cyclin E.
Mutations or dysfunction in NPM1 are linked to diseases such as acute myeloid leukemia and lymphomatoid papulosis. It also participates in fusion proteins (e.g., NPM-ALK, NPM-RAR, NPM-MLF1) associated with tumorigenesis.
