Target Information

STAT3 (Signal Transducer and Activator of Transcription 3) belongs to the STAT protein family, which are phosphorylated by receptor-associated kinases and translocate to the nucleus to act as transcription factors in response to cytokines and growth factors. Coactivators such as CREB-binding protein are required for STAT3 transcriptional activation. STAT3 can be activated by interferons (α, γ), EGF, PDGF, and IL-6. Phosphorylation at tyrosine 705 by JAK1 and JAK2 is critical for dimer formation, nuclear translocation, and DNA binding activity. The STAT3 gene is located on chromosome 17q in humans. It regulates genes involved in cell growth and apoptosis, and knockout of STAT3 in mice is embryonically lethal. STAT3 activity is modulated by Rac1 and inhibited specifically by PIAS3. Three alternatively spliced isoforms of STAT3 have been identified.
