Target Information
Spleen tyrosine kinase (SYK), along with ZAP70, are cytoplasmic tyrosine kinases considered potential drug targets for allergic disorders and autoimmune diseases. SYK may also play a role in cancer. Upon B cell antigen receptor (BCR) or high-affinity IgE receptor (FcεRI) activation, Syk translocates to the plasma membrane and phosphorylates downstream adaptor proteins. This provides docking sites for subsequent signaling pathways, including calcium mobilization, cytoskeleton remodeling, and transcriptional activation of specific genes. Syk binds receptor assemblies through its SH2 domains interacting with phosphorylated ITAM motifs of the receptor, which are phosphorylated by Src-family kinases that also activate Syk via phosphorylation of its activation loop.
