Target Information
This gene encodes a member of the F-box protein family characterized by an approximately 40 amino acid F-box motif. F-box proteins form part of the SCF (SKP1–Cullin–F-box) ubiquitin ligase complex, which mediates phosphorylation-dependent ubiquitination.
F-box proteins are classified into three groups:
- Fbws containing WD-40 domains
- Fbls containing leucine-rich repeats
- Fbxs with other or no recognizable motifs
The encoded BTRC protein belongs to the Fbws class and includes multiple WD-40 repeats. It is homologous to Xenopus bTrCP1, yeast Met30, Neurospora Scon2, and Drosophila Slimb proteins.
BTRC interacts with HIV-1 Vpu, linking CD4 to the proteolytic machinery, and binds phosphorylated IkappaBalpha and beta-catenin destruction motifs. It functions in transcriptional regulation by activating the NF-kappaB pathway and inhibiting the beta-catenin pathway.
