Product Specific Information

The M17/4 monoclonal antibody reacts with mouse CD11a, the 180 kDa integrin alpha L, also known as lymphocyte function-associated antigen-1 (LFA-1) alpha chain. LFA-1, formed by non-covalent association of CD11a with CD18 (integrin beta 2), is a key adhesion molecule involved in lymphocyte and granulocyte function. CD54 (ICAM-1), CD102 (ICAM-2), and CD50 (ICAM-3) are ligands for LFA-1. CD11a is expressed by all leukocytes.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse bone marrow cells. Recommended ≤1 µg/test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number may range from 10⁵–10⁸ cells/test. Optimize by titration for best performance.

Fluorochrome Information:
Super Bright 436 can be excited with the 405 nm violet laser and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

CD11a (LFA-1 alpha, ITGAL) with CD18 forms LFA-1 (alphaLbeta2 integrin), crucial for leukocyte adhesion through ICAM-1, -2, -3 interactions. It plays a role in lymphocyte costimulatory signaling and immune cell activation. CD11a expression shifts from low- to high-affinity conformation upon stimulation, enabling tight ligand binding. Dysfunction in CD11a is associated with immune adhesion disorders. Therapeutic targeting of CD11a (e.g., Efalizumab) has been explored for inflammatory diseases.
