Product Specific Information

Description:
The SK1 monoclonal antibody reacts with the human CD8a molecule, an approximately 32–34 kDa cell surface receptor expressed as a heterodimer with CD8 beta chain (CD8 alpha/beta) or as a homodimer (CD8 alpha/alpha). A majority of thymocytes and a subpopulation of mature T cells and NK cells express CD8a. CD8 binds to MHC class I and, through its association with protein tyrosine kinase p56lck, plays a role in T-cell development and activation.

Applications Reported:
This SK1 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use at 5 µL (0.25 µg) per test, defined for a 100 µL staining volume. Cell number may range from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
Super Bright 436 can be excited with a violet laser (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

Staining Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family, essential for T cell signal transduction, differentiation, and activation.
CD8 is mainly expressed on T cells as a disulfide-linked heterodimer (CD8α/β) functioning as a co-receptor for MHC class I molecules.
CD8 also exists as a homodimer (CD8α/α) on subsets of T cells, NK cells, monocytes, and dendritic cells.
Binding of MHC-I/peptide complexes triggers Lck recruitment, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation.
Activated CTLs play a key role in pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
