Product Specific Information
The PCH101 antibody reacts with the amino terminus of human FOXP3 protein (also known as FORKHEAD BOX P3, SCURFIN, JM2). Cross-reactivity with other proteins has not been determined.
FOXP3, a 49–55 kDa protein, belongs to the forkhead/winged-helix family of transcriptional regulators and is the gene defective in scurfy (sf) mice. Constitutive high expression of FOXP3 mRNA is observed in CD4+CD25+ regulatory T cells (Treg cells), and ectopic expression in CD4+CD25− cells imparts a Treg phenotype.
Intracellular staining of human PBMCs with PCH101 antibody using the anti-human FOXP3 staining set reveals approximately 0.5–4% of lymphocytes staining, predominantly in the CD25^bright population (donor variability dependent).
PCH101 cross-reacts with rhesus, chimpanzee, and cynomolgus species.
Applications Reported: Immunoblotting (WB), immunohistology on frozen and paraffin-embedded tissue sections.
Applications Tested: IHC of formalin-fixed paraffin-embedded tissue using high pH antigen retrieval (20 µg/mL recommended). Antibody should be titrated for optimal assay performance.
Using conjugate solutions: Centrifuge briefly before use and use only the supernatant to minimize nonspecific background staining.
Target Information
FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is 381 amino acids long and highly expressed in CD25+CD4+ regulatory T cells. Defects in FOXP3 cause the scurfy phenotype in mice and IPEX syndrome in humans (immune dysfunction, polyendocrinopathy, enteropathy, X-linked). FOXP3 expression is regulated by p53 in response to DNA damage in human breast and colon cancer cells.
