Product Specific Information
The V9 monoclonal antibody recognizes human Vimentin, a 57 kDa structural protein of intermediate filaments. Vimentin is expressed in mesenchymal-derived cells and specific populations such as radial glia, immature glial cells, and pancreatic precursor cells. It serves as a marker of cardiac differentiation and is gradually replaced by neurofilaments in neural cells. Surface expression has been observed on activated macrophages, platelets, apoptotic T cells, and neutrophils.
This antibody also recognizes canine, rat, and chicken vimentin but not mouse vimentin.
Applications Reported: Western blotting, immunocytochemistry, and immunohistochemistry of frozen (IHC-F) and formalin-fixed paraffin-embedded tissues (IHC-P).
Applications Tested: IHC-P at 1.33 µg/mL using high pH antigen retrieval. Optimal dilution should be determined experimentally.
Using conjugate solutions: Centrifuge briefly before use and apply only the supernatant to reduce nonspecific background staining caused by protein aggregates.
Target Information
Vimentin is a developmentally regulated intermediate filament protein found in cells of mesenchymal origin. It plays a role in intracellular protein transport between the nucleus and plasma membrane. Vimentin is co-expressed with desmin during early development and later replaced by tissue-specific intermediate filaments. It also contributes to steroid synthesis regulation by facilitating cholesterol transport within mitochondria through phosphorylation-induced cytoskeletal changes.
