Target Information

TMP21 is a ubiquitously expressed protein involved in vesicular targeting and protein transport. It is a component of the presenilin complex and modulates the γ-secretase—but not the ε-secretase—cleavage activity of the amyloid precursor protein. This complex includes APH1, nicastrin, PEN2, and presenilin-1, which cleave amyloid precursor protein at γ- and ε-sites, potentially leading to accumulation of Abeta, associated with Alzheimer’s disease. Co-immunoprecipitation experiments have shown TMP21’s association with these proteins, suggesting a regulatory role. Suppression of TMP21 expression by siRNA increases γ-secretase activity and Abeta production, demonstrating TMP21’s modulatory function.
