- Immunohistochemistry (PFA fixed):
Incubate antibody for 30 minutes at room temperature.
For formalin-fixed tissues, heat sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes.
Target Information
The Myogenic determination gene (MyoD) was first identified by its ability to convert embryonic mouse fibroblast cells into muscle cells. Forced expression of MyoD (human homolog: myf3) can induce muscle differentiation or activate muscle-specific genes in otherwise inactive cells.
The regulatory domain of MyoD lies within a 70 amino acid region containing a basic DNA-binding motif and a helix-loop-helix (HLH) dimerization motif.
Related genes sharing sequence homology in the HLH domain include myf5, myogenin (myf4), and myf6 (MRF4, herculin).
