Product Specific Information
The PK136 monoclonal antibody reacts with mouse NK1.1, an antigen expressed by natural killer cells and a subset of T cells in NK1.1 mouse strains including C57BL and NZB.
Common laboratory strains such as BALB/c, SJL, AKR, CBA, C3H, and A do not express NK1.1. For detection of NK cells in these strains, use monoclonal antibody DX5 (Product #14-5971).
Simultaneous staining of C57BL/6 spleen cells with PK136 and DX5 reveals coexpression of both markers by most cells, with small populations of DX5⁺PK136⁻ and DX5⁻PK136⁺ cells.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on C57Bl/6 mouse splenocytes; ≤0.5 µg/test (100 µL final volume).
Cell number: 10⁵–10⁸ cells/test (empirically determined).
Careful titration recommended for optimal performance.
Excitation: 488 nm

Target Information
NK1.1 (Killer cell lectin-like receptor subfamily B, member 1, KLRB1, NKR-P1A, CD161) is expressed by Natural Killer (NK) cells, which mediate cytotoxicity and cytokine secretion after immune stimulation.
KLRB1 protein contains extracellular C-type lectin motifs, a transmembrane domain, and a cytoplasmic domain.
NKR-P1A (CD161), encoded by KLRB1, recognizes Lectin Like Transcript-1 (LLT1) as a functional ligand.
Classified as a type II membrane protein due to its external C terminus.
