Product Specific Information

Description:
The H57-597 monoclonal antibody reacts with the beta chain of mouse TCR. TCR beta is expressed by thymocytes in a developmentally regulated manner and a majority of peripheral T cells. Crosslinking of the TCR complex with H57-597 induces activation and proliferation of T cells or apoptosis depending on assay conditions. It is used as a phenotypic marker for TCR beta expressing T cells.

This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended use ≤ 0.25 µg per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.

Target Information

T cell receptors (TCR) discriminate foreign from self-peptides presented by MHC class II molecules, essential for adaptive immune response. Recognition of self-peptides is linked to autoimmune diseases and tumor-associated mutations. Superantigen engagement can lead to toxic shock syndrome. Autoantibodies to V beta segments of TCRs have been found in rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE), potentially modulating immune responses.
Human T cells mainly express TCR alpha-beta with CD4 or CD8 (SP). Some lack both (DN), and increased alpha-beta DN T cells are observed in autoimmune disorders. Gamma-delta T cells, found in epithelial tissues, show less TCR diversity and distinct antigen recognition, with subsets exhibiting antitumor and immunoregulatory functions.
