Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. SK7 and UCHT1 antibodies cross-block binding, indicating overlapping epitopes.

Applications Tested

• Pre-titrated and tested by flow cytometry using normal human peripheral blood cells
• Recommended use: 5 µL (0.2 µg) per test (100 µL final volume)
• Cell number range: 10⁵–10⁸ cells/test

Important Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
• Add 5 µL of CellBlox Plus Buffer per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits encoded on chromosome 11. CD3 is expressed on most T lymphocytes and thymocytes, but not on B or NK cells. CD3 defects are associated with immunodeficiency.
