Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3ε, a 20 kDa subunit of the TCR complex. The ε chain, along with γ and δ subunits, is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. The SK7 and UCHT1 antibodies cross-block binding, suggesting overlapping epitopes.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for human peripheral blood cells. Use 5 µL (0.4 µg) per test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample prior to mixing with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered to minimize spillover and spectral spread.

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR expression. It includes CD3γ, CD3δ, CD3ε, and CD3ζ subunits, encoded on human chromosome 11. These subunits contain cytoplasmic tails with tyrosine-based motifs mediating signal transduction. CD3 is found on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but absent on B and NK cells. Mutations in CD3 genes are associated with CD3 immunodeficiency.
