Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. The SK7 and UCHT1 monoclonal antibodies cross-block binding, suggesting recognition of overlapping epitopes.

Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells at 5 µL (0.4 µg) per test. A test is defined as staining a cell sample in 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Invitrogen LIVE/DEAD Fixable Dead Cell stains are recommended.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance. Add 5 µL per sample prior to combining with cells. For single-color controls, use 5 µL per 100 µL cell sample containing 10³–10⁸ cells.

Target Information

The CD3 subunit complex transduces antigen-recognition signals into the cytoplasm of T cells and regulates TCR surface expression. CD3 gamma, delta, epsilon, and zeta subunits are encoded by closely linked genes on human chromosome 11. These subunits have cytoplasmic tails that associate with signal transduction molecules via tyrosine-based motifs. CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Defects in CD3 genes are associated with immunodeficiency.
