Product Specific Information

Description:
The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. Along with CD3 gamma and delta, the epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. SK7 and UCHT1 antibodies cross-block binding, suggesting overlapping epitopes.

The antibody SK7 recognizes chimpanzee CD3.
Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported:

• Suitable for flow cytometric analysis.

Applications Tested:

• Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
• Recommended usage: 5 µL (0.2 µg) per test (100 µL final volume).
• Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample or per 100 µL of cell sample (10³–10⁸ cells).
• For antibody cocktails, add CellBlox Plus Blocking Buffer before combining with cells.

Target Information

The CD3 complex is essential for transducing antigen-recognition signals into T cells and regulating TCR surface expression. It includes subunits CD3 gamma, delta, epsilon, and zeta, all encoded on human chromosome 11. These subunits associate with cytoplasmic signaling molecules via tyrosine-based motifs. CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Mutations in CD3 genes are linked to CD3 immunodeficiency.
