Product Specific Information

Description:
The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. Along with other CD3 subunits (gamma and delta), the epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. The SK7 and UCHT1 monoclonal antibodies cross-block binding, suggesting recognition of overlapping epitopes.

Each product contains one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:
This SK7 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
This SK7 antibody has been pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.4 µg) per test in a final volume of 100 µL. Cell number can range from 10⁵ to 10⁸ cells/test.

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

CellBlox Plus Blocking Buffer:
This product ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which provides improved performance over previous formulations. Add 5 µL per sample (regardless of the number of NovaFluors in your panel) for optimal staining. For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

Target Information

The CD3 subunit complex is essential for transducing antigen-recognition signals into T cells and regulating TCR expression. It consists of CD3 gamma, delta, epsilon, and zeta subunits, encoded by closely linked genes on human chromosome 11. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Defects in the CD3 gene are associated with CD3 immunodeficiency.
