Product Specific Information
Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
For formalin-fixed tissues, heat sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes before staining.
Target Information
MAPK14 (p38 alpha)는 스트레스 활성화 단백질 키나아제(MAPK) 계열의 구성원으로, 세포 스트레스, 염증 및 다양한 자극에 대한 반응을 조절합니다. 비인산화 상태에서는 비활성이며, Thr-Gly-Tyr 모티프의 이중 인산화에 의해 활성화됩니다.
p38 MAPK는 네 가지 동형체를 가지며, 조직 발현 및 상위/하위 조절 인자에 대한 친화도가 다릅니다. TNF, IL-1, 열충격 등의 자극 시 MAPK kinase-3/6이 활성화되어 p38 MAPK의 Thr180과 Tyr182를 인산화합니다. 활성화된 p38 MAPK는 ATF-2, CHOP-1, MEF-2 등 여러 전사 인자를 인산화하여 세포 반응을 매개합니다.
