Product Specific Information

The 8C11 monoclonal antibody reacts with human CD325 (N-Cadherin), a 130 kDa member of the Cadherin superfamily composed of five extracellular repeats, a transmembrane domain, and a cytoplasmic domain. CD325-deficient mice exhibit embryonic lethality at day 10 of gestation with major cardiac and neural defects. CD325 is implicated in cardiac development, mesoderm formation, left-right symmetry establishment, and cardiac looping morphogenesis. Its expression is often dysregulated in cancer cells and is associated with cell cycle arrest.

Applications Reported:
Flow cytometric analysis, immunoprecipitation, Western blotting, and immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections.

Applications Tested:

• Flow cytometry: ≤0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test)
• Immunohistochemistry (FFPE): ≤10 µg/mL with low pH antigen retrieval
  Titrate antibody for optimal performance.

Target Information

N-cadherin is a 140 kDa calcium-dependent cell adhesion glycoprotein of the cadherin family. It mediates homotypic cell-cell adhesion and plays a critical role in morphogenetic movements during development. Its function depends on interactions with the actin cytoskeleton via catenin proteins, regulated by β-catenin phosphorylation. N-cadherin consists of five extracellular cadherin repeats, a transmembrane region, and a conserved cytoplasmic tail, essential for gastrulation and left-right asymmetry establishment.
