Target Information

The T cell receptor (TCR) plays a critical role in distinguishing self from non-self peptides presented by MHC class II molecules, enabling an effective adaptive immune response. Recognition of self-peptides by TCRs has been linked to autoimmune diseases, while mutant self-peptides are associated with tumors. Bacterial superantigens can engage TCRs and cause toxic shock syndrome. Autoantibodies against V beta segments of TCRs have been identified in patients with rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE), potentially regulating TH1-mediated inflammatory reactions.

Most human T cells express TCR alpha-beta and either CD4 or CD8 (single positive, SP). A small population lacks both (double negative, DN), which can increase in autoimmune or immunodeficiency conditions. Gamma-delta T cells, found mainly in epithelial tissues, exhibit limited TCR diversity and distinct antigen recognition, contributing to antitumor and immunoregulatory activities.
