Target Information

Lysosomal enzymes containing one or two mannose 6-phosphate (man6P) moieties are transported within the cell by approximately 300 kDa cation-independent mannose 6-phosphate receptors (CI-MPR).
These receptors participate in two interconnected cycles:

1. Biosynthetic cycle: MPR transports newly synthesized lysosomal enzymes from the trans-Golgi network to late endosomes or early lysosomes.
2. Endocytic cycle: Transports extracellular lysosomal enzymes from the plasma membrane via clathrin-coated vesicles to early endosomes.

MPRs cycle between compartments every 3 hours, with most found in late endosomes, fewer in the trans-Golgi network, and about 10% at the cell surface.
The receptor also binds type II insulin-like growth factor (IGF-II) simultaneously with man6P.
A truncated soluble form (~240 kDa) representing the extracellular domain circulates in serum and retains ligand-binding capability.
