Target Information

MDM2 is a ubiquitin ligase for p53 and plays a central role in the regulation of p53 stability. It is located on chromosome 12q. Akt-mediated phosphorylation of MDM2 at Ser166 and Ser186 enhances its interaction with p300, promoting MDM2-mediated ubiquitination and degradation of p53. Phosphorylation also prevents binding to p19ARF, further increasing p53 degradation.

MDM2 negatively regulates p53 by binding and inhibiting its transactivation activity. Overexpression of MDM2 leads to p53 inactivation and reduced tumor suppressor function. Additionally, MDM2 possesses E3 ubiquitin ligase activity, contributing to p53 degradation. It also plays roles in apoptosis and cell cycle regulation and is overexpressed in various human malignancies, including soft tissue carcinomas and breast cancer.

Beyond p53, MDM2 interacts with other proteins such as retinoblastoma 1 and ribosomal protein L5, participating in cell cycle, apoptosis, and tumorigenesis. More than 40 alternatively spliced transcript variants of MDM2 have been identified in both tumor and normal tissues.
