Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. Together with CD3γ and CD3δ, the epsilon chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells.

SK7 and UCHT1 monoclonal antibodies cross-block binding, suggesting overlapping epitopes, whereas OKT3 and SK7 recognize different epitopes. The antibody SK7 also recognizes chimpanzee CD3.

Applications Reported: Suitable for flow cytometric analysis and functional assays.
Applications Tested: Verified by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤0.25 µg/test (100 µL final volume). Cell number may vary from 10⁵–10⁸ cells/test. Titrate for optimal performance.

Storage and Handling: Use in sterile environment.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

The CD3 subunit complex is critical for transducing antigen-recognition signals into T cells and regulating TCR surface expression. Activation through the TCR involves CD3γ, CD3δ, CD3ε, and CD3ζ subunits—members of the immunoglobulin superfamily encoded on chromosome 11.

CD3 subunits possess long cytoplasmic tails with double tyrosine-based motifs that mediate association with signal transduction molecules. CD3 may influence TCR-induced growth arrest, survival, and proliferation. CD3 antigen is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Reduced T lymphocyte percentages are observed in some autoimmune diseases. Defects in CD3 genes are associated with CD3 immunodeficiency.
